Protein aggregation is a common theme underlying many neurodegenerative diseases. Here we demonstrate the application of mid-infrared photothermal (MIP) microscopy for studying secondary structure of protein aggregates in living cells using a Huntington's disease model in yeasts. MIP could spectrally reveal the β-sheet enrichment of protein aggregates in living cells, thus identifying protein aggregates free of perturbation from fluorescent protein tags. We showed a nanometer-scale structural partition of β-sheet core and ɑ-helix shell within the aggregation complex and explored the perturbation from prion Rnq1.
