Morphology-based screening of MSC response to changes in manufacturing conditions can be used to predict the functional effects of many pre-conditioning strategies, including at large scale. This functional prediction extends to MSC-EVs, which demonstrate distinctive modulation of inflammatory microglia in response to changes in priming and scale. Identification of effective priming strategies in large scale manufacturing formats combats the endogenous heterogeneity of clinical translation. Integrative omics characterization of the MSC priming response and corresponding immunomodulatory function may reveal fundamental regulators of priming and contribute to more consistent, efficacious MSC-based therapies.
