(G-264) Formulation & Content Evaluation of Drug Loaded Buccal Films & Nanofibers

Introduction:: The conventional formulation of Acetaminophen is low in efficacy, primarily on account of their first pass metabolism. This study aims to focus on development of Acetaminophen loaded buccal films and nanofibers with twofold objective of offering a rapid as well as a prolonged delivery of acetaminophen coupled with enhanced therapeutic efficacy.

Materials and Methods:: Acetaminophen loaded buccal films and nanofibers were formulated by solvent-casting method and electrospinning method respectively using 12% w/v of polycaprolactone, acetone and 2.4grams of acetaminophen. Buccal films increase the bioavailability of the drug and nanofibers demonstrate potential for effective drug delivery because of their large surface area-to-volume ratios,high porosity ad 3D open porous structures.



Results, Conclusions, and Discussions:: Placebo samples were cut into 1x1 cm² and the films possessing the most desirable physicomechanical properties were selected for further drug content uniformity evaluation using UV-Visible spectrophotometer. Based on these studies, the samples showed more absorbance values in nanofiber samples (2.6 Abs versus 0.679 Abs) and buccal films showed (2.8 Abs versus 0.679 Abs) values when compared to oral suspension. The data observed from this study highlights the feasibility of buccal films and nanofibers as a viable option for loading acetaminophen.
