    Introduction:: The use of adoptive immunotherapy has proven to be a highly effective strategy for treating a variety of human cancers, including melanoma, cervical cancer, lymphoma, and leukemia. To further advance this approach, we present a novel cell culture technology for the expansion of primary human T cells for adoptive immunotherapy. Our approach involves suspending and culturing T cells within microscale alginate hydrogel tubes (AlgTubes) or anchored with the Arg-Gly-Asp (RGD). The cells are suspended in a culture vessel containing a cell culture medium. The objective of this work is to develop a low-cost and automated device that enables scalable and cost-effective production of consistent chimeric antigen receptor (CAR) T cells at the point of care.

    Materials and Methods:: In this study, T cells were cultured in AlgTubes or RGD AlgTubes to assess their growth rate and potency of CD8, CD4, CD3, and immune checkpoint inhibitor proteins. The AlgTube system was compared to traditional 3D static culture methods for these parameters. The cells' potency and protein expression were evaluated by multiple technologies including flow cytometry.

    Results, Conclusions, and Discussions:: Under optimized culture conditions, both the AlgTubes and RGD AlgTubes enable the cultivation of T cells with high cell viability, growth rate, yield, potency, and lower immune checkpoint inhibitor expression. These benefits are highly desirable in the production of CAR-T cells, which require large numbers of high-quality T cells for effective immunotherapy. Therefore, the use of AlgTubes for T cell expansion can potentially improve the effectiveness of CAR-T therapy by providing a more consistent and efficient source of T cells. This novel culture technology also offers the added advantage of reducing culture volume, time, and cost while increasing production, making it a promising tool for point-of-care manufacturing of CAR-T cells.
