An ototopical formulation has been developed to achieve successful single-dose treatments of acute otitis media in chinchilla models. The formulation contained the thermo-responsive P407 hydrogel, an antibiotic, and a repurposed transtympanic chemical permeation enhancer (CPE) for non-invasive drug delivery. This formulation is more simplified compared to prior hydrogels with combinatorial CPEs. Additionally, carefully selection of the CPE was able to tune the interactions among the CPE, drug, and P407, resulting in improved hydrogel properties and higher drug flux across the biological tissue.
