    Introduction:: Therapeutic humanized monoclonal antibodies (mAbs) are currently the most effective and expected medicines for a wide range of diseases because of their specificity and efficiency. The therapeutic activity of mAbs is closely related to its concentration in serum as in many pharmaceuticals, and it is known that inappropriate concentrations of therapeutic humanized mAbs may lead to acquiring drug resistance for cancer cells and unexpected side effects, therefore the precise monitoring of therapeutic humanized mAbs is strongly needed. However, it is difficult to analyze the therapeutic humanized mAbs from human samples because of their structure. The target humanized mAb only differs from the complementarity determining region (CDR) from the abundantly existing IgGs. Currently available methods to detect the therapeutic humanized mAbs require a long time for detection, complex manipulation, and large equipment which is operated by specialists, therefore, the development of methods/devices applicable for point-of-care testing (POCT) and furthermore continuous monitoring are in great demand. Recently, we reported a novel biosensing system for a representative humanized therapeutic mAb, bevacizumab, employing anti-idiotype aptamers [1][2]. In this study, we further investigate developing disposable sensors using anti-idiotype biosensing molecules for POCT, using the anti-idiotype biosensing molecules, which recognize the CDR of bevacizumab.

    Materials and Methods:: We developed an electrochemical sensor for bevacizumab using anti-idiotype biosensing molecules, the antigen VEGF and the anti-idiotype aptamer. These anti-idiotype biosensing molecules immobilized electrodes were used for the bevacizumab detection by electrochemical impedance spectroscopy (EIS). We characterized the fabricated sensors both in a PBS solution and in an artificial serum solution to detect the bevacizumab concentration.

    Results, Conclusions, and Discussions:: In gold rod electrodes, by the addition of several concentrations of bevacizumab, the Nyquist plot change was observed depending on the target concentration in both anti-idiotype biosensing molecules. The correlation of peak current change and logarithmic values of bevacizumab concentration was observed with a dynamic range of 10-10000 pM. The obtained limit of detection (LOD) of antigen VEGF and anti-idiotype aptamer was 3.2 pM and 5.9 pM, respectively. We will present the results with the disposable electrodes toward the POCT sensing of bevacizumab.
