(D-154) Engineering Novel Dual Decoy Receptor Antagonists for Neovascular Eye Diseases

Introduction:: Neovascularization in the eye drives the pathogenesis of diseases including age-related macular degeneration and diabetic retinopathy, two of the leading causes of blindness in adults. Vascular endothelial growth factor (VEGF) ligands are major stimulants of neovascularization, and suppression of VEGF ligands binding with their cognate receptors (VEGFRs) on responsive cells has shown clinical benefit in treatment of neovascular eye diseases. Current FDA-approved therapies primarily target VEGF-A, the most prominent regulator of blood vessel formation, in order to prevent it from binding to and activating signaling through VEGFR-1 and VEGFR-2. However, the VEGF-C ligand has been increasingly implicated in pathological angiogenesis, and is found to be upregulated following anti-VEGF-A treatment. Moreover, VEGF-C is as potent as VEGF-A in driving retinal neovascularization under hypoxic conditions. No current therapies bind to both VEGF-A and VEGF-C, which would provide a more complete blockade of neovascularization. We designed novel dual decoy receptor antagonists by fusing the ligand-binding domains of VEGFR-1 and VEGFR-2 to a human antibody Fc domain to allow for high-affinity inhibition of both VEGF-A and VEGF-C and coordinate a more comprehensive blockade of pro-angiogenic signaling pathways. We anticipate that this concurrent blockade of VEGF-A and VEGF-C could lead to enhanced therapeutic efficacy in the treatment of neovascular eye disease.

Materials and Methods:: VEGF-A and VEGF-C dimers as well as dual decoy receptor antagonists were produced in a HEK293F mammalian cell expression system and purified using Ni-NTA or Protein G resin before undergoing size-exclusion chromatography on an FPLC instrument. Bio-layer interferometry (BLI) binding studies were performed using an OctetRED96 instrument. Biotinylated VEGF-A or VEGF-C was immobilized on streptavidin-coated biosensors, and these sensors were then dipped into wells containing dilutions of antagonist or antagonist plus preincubated VEGF ligand for simultaneous binding studies. Signaling studies are being conducted via western blotting, and proliferation, survival, and migration analyses are being performed using the XCelligence platform.


Results, Conclusions, and Discussions:: Results and Discussion: Dual decoy receptor antagonists were found to express well without significant aggregation in the HEK293F mammalian cell expression system. BLI studies demonstrated that these antagonists bound VEGF-A, VEGF-C, and PlGF-1 with high affinity, and also could simultaneously engage both VEGF-A and VEGF-C ligands (Figure 1). In vitro functional characterization studies of the dual decoy receptor antagonists are currently ongoing, including western blot analysis of multiple intracellular angiogenic signaling pathways as well as phenotypic readouts of proliferation, survival, and migration in human umbilical vein endothelial cells (HUVECs). In vivo studies are also currently underway investigating gene delivery of our antagonists as a strategy to block angiogenesis to treat neovascular eye diseases.


Conclusion: We have designed and characterized high-affinity protein antagonists that can engage both VEGF-A and VEGF-C simultaneously. These proteins are being further characterized for in vitro and in vivo efficacy with the goal of further developing these molecules as translational therapeutics to treat neovascular eye diseases.
